[주성분코드:167502ATB 에 따른 심사지침열람]
성인 : 그리세오풀빈으로서 1일 250-500mg(역가)을 1-수회 분할 경구 투여한다. 증상에 따라 최초 1000mg(역가)을 투여하고 효과가 있으면 250-500mg(역가)으로 감량 한다.
어린이 : 1일 체중 kg당 10mg (역가)을 1-수회 분할 경구투여한다.

{체중 25kg 1일 250-500mg(역가)투여}
연령, 증상에 따라 적절히 증감한다.

- 포르피린증 환자
- 간장애(간세포부전)환자
- 이 약에 과민증의 병력이 있는 환자
- 홍반성 루푸스(LE) 환자
- 다형 삼출성 홍반 환자
- 표피괴사 환자
- 임부 또는 임신하고 있을 가능성이 있는 부인

| 이상반응 |
- 과민증 :
과민증 드물게 혈관신경성 부종, 광감수성이 나타날 수 있으므로 이러한 경우에는 투여를 중지한다.
피부 : 드물게 리엘증후군(중독성표피괴사증) 때때로 발진, 홍반, 전신성 홍반성 루푸스(SLE), 다형 삼출성 홍반, 표피괴사, 두드러기, 박탈성 피부염, 색소침착 등의 증상이 일어날수 있으므로 충분히 관찰하고 이상이 인정되는 경우는 중지하는 등 적절한 처치를 한다.
전신성 홍반성 루프스(SLE)양 증상 : 드물게 SLE양 증상(발열, 홍반, 근육통, 관절통, 림프절, 옹종등)이 나타날 수 있다.
혈액 : 때때로 백혈구 감소, 과립구 감소, 백혈구증가 등이 나타날 수 있다.
간장 : 때때로 GOT의 증가, BSP 배설지연 등이 나타날수 있다.
소화기 : 때때로 식욕부진, 구역, 위장장애, 아구창, 구토, 설사, 상복부불쾌감 드물게 미각이상, 설통, 구갈, 위장관 출혈 등이 나타날 수 있다.
정신신경계 : 때때로 현기, 두통, 두중, 졸음, 불면, 피로 드물게 이명, 정신착란, 우울상태, 시력장애, 말초신경염 등이 나타날 수 있으며, 장기 투여시 손과 발의 지각이상이 일어날 수 있다.
신장 : 때때로 단백뇨, 신증 등이 나타날 수 있다.
대사이상 : 드물게 포르피린증이 나타날 수 있다.
기타 : 때때로 권태감, 발열, 관절통, 드물게 여성형 유방, 월경이상이 나타날 수 있다.

| 상호작용 |
- 이 약은 알코올의 작용을 증강하고 또 쿠마린계(와르파린계)항응혈의 작용을 약화하므로 이들과 병용투여시 용량에 주의한다.
- 바르비탈산 유도체와의 병용투여시 이 약의 작용이 저하되므로 용량에 주의한다.
- 경구용 황체.난포 호르몬과의 병용투여시 그 작용을 감소시킨다는 보고가 있다.

Griseofulvin에 대한 Mechanism_Of_Action 정보
Griseofulvin is fungistatic, however the exact mechanism by which it inhibits the growth of dermatophytes is not clear. It is thought to inhibit fungal cell mitosis and nuclear acid synthesis. It also binds to and interferes with the function of spindle and cytoplasmic microtubules by binding to alpha and beta tubulin.

Griseofulvin is a mycotoxic metabolic product of Penicillium spp. It was the first available oral agent for the treatment of dermatophytoses and has now been used for more than forty years. Griseofulvin is fungistatic with in vitro activity against various species of Microsporum Epidermophyton, and Trichophyton. It has no effect on bacteria or on other genera of fungi. Following oral administration, griseofulvin is deposited in the keratin precursor cells and has a greater affinity for diseased tissue. The drug is tightly bound to the new keratin which becomes highly resistant to fungal invasions.
